Multichain structure of the IFN-alpha receptor on hematopoietic cells.

摘要: The structure of IFN-alpha receptor was studied by 1) developing antibodies against the receptor, and 2) screening a number cell lines affinity cross-linking to identify cells that express different 2 structures. We report two patterns are observed in human hematopoietic origin. predominant form is multichain which forms complexes 110 130 kDa (alpha-subunit). A high Mr complex 210 results from association alpha-subunit other components. In contrast, another has been identified IFN-alpha-resistant U-937 line some cases acute leukemia. This characterized presence alpha- subunit, absence 110- 210-kDa bands. Also novel 180-kDa more prominent 75-kDa band observed. Functional studies performed showed this not only partially resistant antiproliferative antiviral effects IFN-alpha, but also fails down-regulate upon binding.
