摘要: Unlike TCR-alpha beta cells, TCR-gamma delta cells express a distinct member of the zeta family, gamma-chain Fc epsilon RI (Fc gamma) within TCR complex. To study role in transgenic mouse (G8) has been crossed with CD3 zeta-chain-deficient mice (G8.zeta-/-). Thy-1+ spleen and lymph node these animals expressed low levels CD3/TCR. These results suggested that zeta-chain is required for effective transport to cell surface. In contrast, intraepithelial G8.zeta-/- high TCR. Immunoprecipitation anti-CD3 showed gamma-chains were associated complex T isolated from zeta-deficient mice. Although gamma-expressing proliferated response stimulation by TCR-specific Abs including epsilon, anti-pan gamma delta, anti-V 2 mAb, did not respond G8-specific Ag (T10b), anti-Thy-1 or Con A. The unresponsiveness was due reduced expression, because Ag. inability could be overcome providing an anti-CD28 costimulatory signal adding exogenous rIL-2. Taken together, our data suggest associates absence zeta-chain, but it able substitute surface functional responses
