Transplantation of olfactory ensheathing cells combined with chitosan down-regulates the expression of P2X7 receptor in the spinal cord and inhibits neuropathic pain

摘要: Abstract Background Neuropathic pain (NPP) is the common symptom of most clinical diseases, and its treatment has always been a difficult problem at present. Therefore, purpose this study to explore new method for NPP by transplanting olfactory ensheathing cells combined with chitosan (OECs-CS). Methods Animal model chronic compression sciatic nerve injury (CCI) was made, (OECs) were cultured, (CS) biomaterials prepared, biocompatibility OECs CS detected MTT method, OECs-CS transplanted into site injured respectively, behavioral used measured mechanical withdrawal thresholds (MWT) thermal latency (TWL) rats. On days 7 14 after surgery, expression level P2X7 receptor (P2X7R) in L4-5 spinal cord using situ hybridization, western-blotting qRT-PCR. To therapeutic effect transplantation on suppression. Results After injury, MWT TWL rats significantly reduced, levels P2X7R protein mRNA increased. OECs-CS, Importantly, compared OECs, could better reduce P2X7R, relieve hyperalgesia Moreover, CCI + OECs-CS group reduced relieved. Conclusion can inhibit overexpression mediated NPP, while than alone. Our results provide novel theoretical basis NPP.
