摘要: Abstract Chlorpromazine is used in the treatment of schizophrenia and psychotic disorders belongs to phenothiazine class neuroleptic drugs. It shows severe side effects such as extrapyramidal symptoms well ocular skin disorders, but mechanism still not fully established. The aim this study was examine effect chlorpromazine on cell viability, melanogenesis antioxidant defense system normal human melanocytes. has been demonstrated that induces concentration-dependent loss viability. value EC 50 calculated be 2.53 μM. lower concentrations (0.0001, 0.001 0.01 μM) increased melanin microphthalmia-associated transcription factor (MITF) content tyrosinase activity, while changes enzymes activity were observed. suggests long-term therapy, even with low drug doses, may lead hyperpigmentation and/or eye. use analyzed higher (0.1 1.0 μM) caused significant alterations melanocytes, what explain a potential role depletion cellular status leading other adverse associated high-dose therapy.
