摘要: The beneficial effects of electroacupuncture (EA) at Shuigou (GV26) and Neiguan (PC6) on poststroke rehabilitation are critically related to the activation delta-opioid receptor (DOR). underlying anti-inflammatory mechanisms in DOR EA-mediated neuroprotection cerebral ischemia/reperfusion (I/R) injury were investigated current study. Cell proliferation apoptosis detected by morphological changes, cell counting kit-8 (CCK-8) assay, lactate dehydrogenase (LDH) release, TUNEL staining. mRNA levels evaluated using real-time quantitative polymerase chain reaction (RT-qPCR), protein expression was measured western blot or enzyme-linked immunosorbent assay (ELISA) vitro. Infarct volume examined cresyl violet (CV) staining, neurologic recovery assessed neurological deficit scores, pro- cytokines determined immunofluorescence vivo. greatly ameliorated injury, reduced LDH leakage apoptosis, increased viability. It reversed oxygen-glucose deprivation/reoxygenation- (OGD/R-) induced downregulation protein, as well BDNF protein. also proinflammatory cytokine gene expression, including TNF-α, IL-1β, IL-6, same time, IL-4 IL-10 OGD/R challenged PC12 cells. EA significantly middle artery occlusion/reperfusion- (MCAO/R-) infarct attenuated scores. markedly decreased while sham did not have any protective effect MCAO/R-injured rats. plays an important role against inhibiting inflammation via brain-derived neurotrophic factor/tropomyosin-related kinase B (BDNF/TrkB) pathway. neuroprotective efficacy I/R may be inhibition inflammatory response through DOR-BDNF/TrkB
