Severe In Vivo Hyper-Homocysteinemia is not Associated with Elevation of Amyloid-β Peptides in the Tg2576 Mice

摘要: Since hyper-homocysteinemia (HHcy) was recognized as a risk factor for Alzheimer's disease (AD), many studies tried to induce HHcy in animal models investigate its effect on amyloid-beta protein precursor (AbetaPP) metabolism. Previous reports found that induced AD transgenic mouse models, by either feedina methionine-enriched diet or vitamin Bs deficient diet, is associated with elevation of (Abeta) levels. However, there no data available the dietary intervention which combines both excessive methionine and low levels amyloidogenesis any these models. In current study, we investigated combination enriched folate, B6 B12, an model, Tg2576. We 7 months treatment this severe mice plasma homocysteine level higher than 150 microM. difference detected brain Abeta deposition between diet-treated control group. As shown western blot, did not alter steady state proteins involved AbetaPP metabolism, either. These results demonstrate diet-induced does influence vivo.
