O2A progenitor cells transplanted into the neonatal rat brain develop into oligodendrocytes but not astrocytes

摘要: Abstract The differentiation of the bipotential O2A progenitor cell into an oligodendrocyte or a type 2 astrocyte has been well documented in cultures various regions central nervous system. The appropriate tools to prove its existence vivo have lacking. We report on vitro-in approach that combines stable labeling enriched population cultured progenitors by fluorescent dye fast blue, followed their transplantation neonatal rat brains, which allowed us study influence brain microenvironment lineage decision. grafted cells survived and 21 days after grafting nearly all were positive for oligodendroglial marker galactocerebroside. Surprisingly, blue-positive did not stain astroglial glial fibrillary acidic protein. These results indicate progenitor's plasticity is restricted environment, resulting developmental exclusion initially described vitro.
