Rotenone-induced apoptosis is mediated by p38 and JNK MAP kinases in human dopaminergic SH-SY5Y cells.

摘要: Rotenone is a naturally derived pesticide that has recently been shown to evoke the behavioral and pathological symptoms of Parkinson's disease in animal models. Though rotenone known be an inhibitor mitochondrial complex I electron transport chain, little about downstream pathways leading its toxicity. We used human dopaminergic SH-SY5Y cells study mechanisms rotenone-induced neuronal cell death. Our results suggest rotenone, at nanomolar concentrations, induces apoptosis caspase-dependent. Furthermore, treatment phosphorylation c-Jun, c-Jun N-terminal protein kinase (JNK), p38 mitogen activated (MAP) kinase, indicative activation JNK pathways. Importantly, expression dominant interfering constructs or attenuated apoptosis. These data requires MAP kinases caspases. studies provide insights concerning molecular cells.
