摘要: The relationships between gene expression, protein and enzymatic activity of thymidylate synthase (TS) dihydropyrimidine dehydrogenase (DPD) have not been clarified in tumor non-tumor tissues human. In this study, we compared three different parameters by evaluating TS DPD levels, mRNA expression assessed RT-PCR, evaluated immunohistochemical examination, measured biochemical assay, the tissue adjacent normal mucosa 43 patients with gastrointestinal cancer. was significantly higher than both stomach (median activity: 81.0 38.0 pmol/mg protein, respectively, p=0.012) colorectum (49.8 30.8, p=0.023). Similarly, TS/GAPDH ratio: 6.0 2.0, p=0.009) (3.20 0.91, p=0.001). But no significant differences were observed either 41.3 41.6 pmol/ min/mg respectively) or (34.9 49.0, respectively). On other hand, levels only (DPD/GAPDH 0.83 0.20, p=0.003). No linear found tissue. Nor any correlations for DPD. These results suggest that may vary methods used to measure them. discrepancies must be taken into account when interpreting correlation clinical outcome.
