摘要: The structures of the asparagine-linked oligosaccharide chains IgG from autoimmune arthritic MRL/Mp-lpr/lpr (MRL-lpr/lpr) mice and control MRL/Mp(-)+/+ (MRL(-)+/+) were investigated. Two subpopulations IgG, M1-I M1-II, obtained serum MRL-lpr/lpr by column chromatography on protein A-Sepharose CL-4B. Although did not bind to column, its elution was retarded, whereas M1-II bound eluted in acidic buffer. (Mn) MRL(-)+/+ showed same chromatographic behavior as M1-II. liberated quantitatively hydrazinolysis samples Mn, M1-I, a pooled mixture (M1) determined sequential exoglycosidase digestion, lectin (RCA120) affinity HPLC, methylation analysis. Their shown be biantennary complex-type +/- Gal beta 1----4GlcNAc 1----2Man alpha 1----6(+/- 1----3)Man 1----4(+/- Fuc 1----6)GlcNAc. proportion each Mn but differed that where degree galactosylation significantly decreased which caused change pattern total mice. This phenomenon, is also found patients with rheumatoid arthritis, suggests alteration oligosaccharides may common feature animals develop arthritis production factor regardless species.
