摘要: The radioallergosorbent test (RAST) was described in 1967 by Wide, Johansson and Bennich as an vitro diagnostic for allergen antibodies [1]. Figure 1 shows the principle of RAST. Allergens are covalently bound to solid-phase Polysaccharides which have been activated reaction with cyanogen bromide. In first step RAST reacts serum, allergens including IgE bind. second RAST, after washing remove unbound antibodies, allergen-antibody complex radioiodinated affinity chromatography-purified antibody IgE. This is washed again, radioactivity measured. indicates that quantity allergen-IgE proportional step. original procedure utilized particles suspension, such Sephadex or cellulose. Subsequently, commercial version has simplified use paper discs; whereas suspensions require centrifugation washing, disc can be fluid simply aspirated. a considerable stimulus investigation diseases associated permitting quantitative measurement levels [2].
