Development of novel transcriptome-based SSR markers in Puccinia triticina and their potential application in genetic diversity studies

摘要: In wheat, leaf rust caused by Puccinia triticina Eriks is a major disease that causes losses in production. the present study, microsatellite or SSR (simple sequence repeat) markers were developed for P. using transcriptome data obtained during wheat-P. interactions. A total of 37,268 differentially expressed transcripts comparative transcriptomics and these, 6809 (1.8%) carried SSRs. Tri-nucleotide SSRs most abundant (52%) among identified Primer pairs synthesized 50 PtESSRs, which 28 found to be polymorphic pathotypes Indian subcontinent. The number alleles per locus ranged from 2 12 with an average 5.72. Polymorphic information content (PIC) 0.29 0.96 0.69. observed heterozygosity 0.10 0.48 0.21. Interestingly, one marker, namely PtESSR7, could distinguish between different virulence groups pathotypes. BLAST analysis silico prediction fungal effectors suggested some PtESSR may have potential functional pathology. Dendrogram analyses detected two clusters 48 Results this study suggest newly are useful genetic studies pathogen. No relationship was diversity geographic distributions populations.
