Characterization of the sortase A from Lactobacillus acidophilus ATCC 4356 involved in adherence to intestinal cells.

摘要: Aim: Confirmation of the enzymatic activity Class A sortase (SrtA) in probiotic strain Lactobacillus acidophilus associated with adhesion properties. Materials & methods: SrtA from L. ATCC 4356 was purified and its properties investigated by site-directed mutagenesis approach sensitivity to metal ions also detected. Results: ATCC4356 can recognize LPxTG LPxTD sorting motifs. The active sites include His137, Cys198 Arg205. Furthermore, acacetin increase SrtA, while phenyl vinyl sulfone could effectively inhibit an IC50 143.32 μg/ml. ability decreased resulting inhibition activity. Conclusion: unique provide some insights into development high-adhesion strains GI tract.
