摘要: Rapid separation of large numbers human peripheral blood mononuclear cells into fractions enriched for B lymphocytes, T or monocytes was accomplished by counterflow centrifugal elutriation (CCE). The first fraction contained 98% the platelets. Ten additional containing subpopulations were collected sequential increases in flow rate while maintaining a constant centrifuge speed. Analysis using monoclonal antibodies revealed that 2, which free esterase-positive monocytes, highly cells. lymphocytes (OKT3+) predominant cell type found 4. No enrichment T-lymphocyte-helper (OKT4+) -suppressor (OKT8+) observed lymphocyte fractions. Three (7-9), cells, predominantly OKM1+ with no evidence selective monocyte subpopulations. Thus, and could be obtained, utilizing CCE, enough quantities to enable analysis their functional properties. Of particular interest ability separate small, resting from monocytes.
