MEK1 dependent and independent ERK activation regulates IL-10 and IL-12 production in bone marrow derived macrophages

摘要: The mitogen activated protein kinases ERK1/2 play an important role in response to toll like receptor (TLR) activation and cytokine production, including IL-10 IL-12. Here, we examined the of MEK1 TLR4 agonist by using bone marrow-derived macrophages (BMDMs) from wild type (WT) Mek1(d/d)Sox2(Cre) mice. Our data demonstrates that is essential for LPS. Furthermore, stimulation BMDMs derived mice showed enhanced STAT4 phosphorylation increased IL-12 secretion, but exhibited a significantly lower production as compared WT macrophages. Most interestingly, TLR ligation presence recombinant (rIL-10) or retinoic acid (RA) led independent inhibition decreased levels. Collectively, these suggest required mediated ERK turn regulates It also indicates can be RA this negatively IL-12, positively production. These findings may have significant implications development drugs modulate activity treatment inflammatory, autoimmune proliferative diseases such cancer.
