The excitation–contraction coupling mechanism in skeletal muscle

摘要: First coined by Alexander Sandow in 1952, the term excitation–contraction coupling (ECC) describes rapid communication between electrical events occurring plasma membrane of skeletal muscle fibres and Ca2+ release from SR, which leads to contraction. The sequence twitch involves: (1) initiation propagation an action potential along membrane, (2) spread throughout transverse tubule system (T-tubule system), (3) dihydropyridine receptors (DHPR)-mediated detection changes potential, (4) allosteric interaction DHPR sarcoplasmic reticulum (SR) ryanodine (RyR), (5) SR transient increase concentration myoplasm, (6) activation myoplasmic buffering contractile apparatus, followed (7) disappearance myoplasm mediated mainly its reuptake through adenosine triphosphatase (SERCA), under several conditions movement mitochondria extrusion Na+/Ca2+ exchanger (NCX). In this text, we review basics ECC techniques used study it. Moreover, highlight some recent advances point out gaps knowledge on particular issues related such as DHPR-RyR molecular interaction, differences regarding fibre types, alteration during fatigue, role store-operated entry general sequence, potentiators, sparks.
