摘要: The transport, accumulation, and distribution of ascorbic acid were investigated in isolated human neutrophils utilizing a new assay, which combined the techniques high performance liquid chromatography coulometric electrochemical detection. Freshly contained 1.0-1.4 mM acid, was localized greater than or equal to 94% cytosol, not protein bound, present only as dehydroascorbic acid. Upon addition extracellular medium physiologic amounts, accumulated millimolar concentrations. Accumulation mediated by affinity low transporter; both transporters responsible for maintenance concentration gradients large 50-fold. transporter had an apparent Km 2-5 microns Lineweaver-Burk Eadie-Hofstee analyses, 6-7 similar analyses. Each saturable temperature dependent. In normal blood should be saturated, whereas its linear phase uptake.
