Excitation of locus coeruleus neurons by vasoactive intestinal peptide: role of a cAMP and protein kinase A.

摘要: In accord with previous studies, it was found that vasoactive intestinal peptide (VIP), a powerful activator of adenylate cyclase, and cAMP-active agents (i.e., 8-Br-cAMP, forskolin, Ro20-1724) increased the firing rate noradrenergic neurons in locus coeruleus (LC) by inducing an inward current. The response to VIP usually more rapid larger subpopulation LC subthreshold rhythmic oscillations membrane potential (oscillatory cells) as compared nonoscillatory cells. either case, currents elicited were be nonadditive, suggesting action VIP, at least part, is via same mechanism agents. Intracellular application specific protein (or related peptide) inhibitor cAMP-dependent kinase markedly attenuated activation induced or (protein A), presumably through phosphorylation, plays role VIP. Taken together, results provide evidence cAMP A are involved mediating electrophysiological actions on neurons.
