Intragenic recombination influences rotavirus diversity and evolution.

摘要: Because of their replication mode and segmented dsRNA genome, homologous recombination is assumed to be rare in the rotaviruses. We analyzed 23,627 complete rotavirus genome sequences available NCBI Virus Variation database, found 109 instances recombination, at least eleven which prevailed across multiple sequenced isolates. In one case, may have generated a novel VP1 lineage. also strong evidence for intergenotypic more than sequence strongly supported same event, particularly between different genotypes segment 9, encodes glycoprotein, VP7. The recombined regions many putative recombinants showed amino acid substitutions differentiating them from major minor parents. This finding suggests that these events were not overly deleterious, since presumably proliferated long enough acquire adaptive mutations regions. Protein structural predictions indicated that, despite sometimes substantial replacements resulting overall protein structures remained relatively unaffected. Notably, junctions appear occur nonrandomly with hot spots corresponding secondary RNA structures, pattern seen consistently segments. total, we nine A Only segments 7 (NSP3) 11 (NSP5) did show recombination. Collectively, results our computational analyses suggest contrary prevailing sentiment, significant driver evolution influence circulating strain diversity.
