摘要: The effect of the extracellular chaperone, clusterin, on amyloid fibril formation by lipid-free human apolipoprotein C-II (apoC-II) was investigated. Sub-stoichiometric levels derived from either plasma or semen, potently inhibit apoC-II. Inhibition is dependent apoC-II concentration, with more effective inhibition clusterin observed at lower concentrations average sedimentation coefficient fibrils formed (0.3 mg.mL-1) reduced coincubation (10 microg x mL(-1)). In contrast, addition (0.1 mg mL(-1)) to preformed does not affect size distribution after 2 days. This velocity data suggests that inhibits growth but promote dissociation. Electron micrographs indicate similar morphologies for in presence absence clusterin. substoichiometric nature interacts transient nuclei leading dissociation monomeric subunits. We propose a general role suppressing amyloid.
