摘要: This study describes the features of tissue-engineering scaffold capable sequentially delivering three angiogenic factors. The consists alginate-sulfate/alginate, wherein vascular endothelial growth factor (VEGF) platelet-derived factor-BB (PDGF-BB) and transforming factor-beta1 (TGF-beta1) are bound to alginate-sulfate with an affinity similar that realized upon their binding heparin. Factor release rate from was correlated equilibrium constants factors matrix, thus enabling sequential delivery VEGF, PDGF-BB TGF-beta1. In alginate scaffolds lacking alginate-sulfate, adsorbed proteins instantaneous. After subcutaneous implantation for 1 3 months in rats, blood vessel density percentage mature vessels were 3-fold greater triple factor-bound than factor-adsorbed or untreated scaffolds. Moreover, vascularization within superior found only basic fibroblast factor. Application this novel may be extended combined additional heparin-binding combinations active different tissue regeneration processes.
