Characterization of extended-spectrum-β-lactamase producing Klebsiella pneumoniae phage KP1801 and evaluation of therapeutic efficacy in vitro and in vivo.

摘要: Extended spectrum β lactamase-producing Klebsiella pneumoniae (ESBL-KP) is being reported with high morbidity and mortality rates considered as the highest priority for new antimicrobial strategies. To develop an alternative agent, phage KP1801 broad lytic activity was isolated. The genome of double stranded DNA 49,835 base pairs, a GC content 50.26%. There were 75 putative open reading frames. Phage classified in the order Caudovirales, belonging to Siphoviridae family. About 323 proteins detected by shotgun proteome analysis. inhibited biofilm formation reduced pre-formed dose dependent manner. Scanning electron microscopic studies demonstrated membrane damage bacterial cells treated phage, resulting cell death. Prophylactic therapeutic efficacies evaluated Galleria mellonella. Administration ESBL-KP infection significantly improved survival G. number intracellular bacteria larvae showed significant decrease compared untreated control while increased. These suggested that has potential development antibiotics biocontrol agents against infection.
