High-density multi-fiber photometry for studying large-scale brain circuit dynamics

摘要: Animal behavior originates from neuronal activity distributed and coordinated across brain-wide networks. However, techniques to assess large-scale brain circuit dynamics in behaving animals remain limited. Here we present compact, high-density arrays of optical fibers that can be chronically implanted into the mammalian brain, enabling multi-fiber photometry as well optogenetic perturbations many regions. In mice engaged a texture discrimination task achieved simultaneous photometric calcium recordings networks 12 48 regions, including striatal, thalamic, hippocampal, cortical areas. Furthermore, optically perturbed specific subsets regions VGAT-ChR2 by using spatial light modulator address respective fiber channels. Perturbation ventral thalamic nuclei caused network modulation behavioral deficits. Finally, demonstrate freely moving animals, two during social interaction. Thus, are simple, low-cost, versatile tools open novel ways investigate behavior.
