摘要: Colon cancer is one of the deadliest cancers worldwide; abnormal microRNA expression common during colon development. The aim present study was to elucidate role played by miR-185 in this context. We used quantitative real-time PCR (qRT-PCR) measure levels cell lines. effects on proliferation and invasion were assessed using MTT, colony-forming, wound-healing, transwell assays. A luciferase activity assay confirm target miR-185. Our data showed that significantly down-regulated cells colonic tissues compared with NCM460 normal epithelial adjacent tissues. functional analysis revealed ectopic inhibited proliferation, colony formation, migration, invasion. In addition, western blot, qRT-PCR, assays confirmed Wnt1 a downstream miR-185, turn suppressing β-catenin-mediated signaling. conclusion, we found inhibits targeting Wnt1, it serves as tumor suppressor, indicating modulation may potentially be therapeutic patients.
