摘要: Trout primary kidney monocyte-like cultures (T-PKM) were generated by incubating head leukocytes in the presence of cell-conditioned medium (CCM). This technique was adapted from procedures that previously used to cultivate vitro-derived macrophages (IVDKM) goldfish. Flow cytometric analysis initial T-PKM cultures, identified three cell sub-populations, but only one these sub-populations survived extensive cultivation periods (i.e. >8 days) CCM. Functionally, reactive oxygen intermediate (ROI) production detected following stimulation with PMA. However, cells failed produce nitrogen intermediates (RNI) response immunological stimuli. In contrast, goldfish IVDKM capable producing both ROI and RNI. Using dihydrorhodamine (DHR) assay flow cytometry, we two ROI-producing a single sub-population present after extended T-PKM. subsequently sorted using cytometer shown possess morphology microscopic analysis.
