摘要: The permeability of the yeast cells (Saccharomyces cerevisiae) to lipophilic tetraphenylphosphonium cations (TPP(+) ) after their treatment with single square-shaped strong electric field pulses was analyzed. Pulsed fields (PEF) durations from 5 150 µs and strengths 0 10 kV/cm were applied a standard electroporation cuvette filled appropriate buffer. TPP(+) absorption process analyzed using an ion selective microelectrode (ISE) plasma membrane determined by measurements obtained calcein blue dye release assay. viability inactivation optical absorbance method. experimental data taken yeasts treated PEF incubated for 3 min showed increased uptake yeast. This can be controlled setting amplitude pulse duration PEF. kinetics is described second order absolute rate equation. It concluded that changes charge on cell wall, which main barrier , due poration membrane. applicability analysis also discussed.
