Action of the stable prostacyclin analogue iloprost on microvascular tone and -permeability in the hamster cheek pouch

摘要: In order to further elucidate the mechanisms involved in therapeutic effects of prostacyclin and Iloprost peripheral ischemic disease, actions on microvascular tone, capillary density, increases venular permeability induced by inflammatory mediators ischemia were investigated cheek pouch anaesthetized Syrian hamsters using intravital videomicroscopy - for quantification vascular leakage fluorescein-labelled dextran (FITC-D; Mw 70,000). at nonhypotensive, platelet aggregation-inhibiting dose 0.5 μg/kg/min i.v. significantly increased diameters arterioles venules density perfused capillaries antagonized vasoconstriction decrease as Leukotriene D4 (LTD4; 10−7 M). FITC-D histamine (10−5 M), serotonin bradykinin (10−6 M) reperfusion after 30 min ischemia. Topical application (10−8 intraarterial infusion Prostaglandin E1 (PGE1; 2.0 μg/kg/min), topical Forskolin also attenuated histamine-induced leakage, while PGE1 (10−7M) Nifedipine (30 μg/kg + 10 μg/kg/min) not effective. It is concluded, that improvement tissue perfusion functional antagonism mediator-induced edema vasospasm could contribute effectiveness diseases.
