DNA Polymorphisms in the Sahiwal Breed of Zebu Cattle Revealed by Synthetic Oligonucleotide Probes

摘要: Genomic DNA of 15 randomly selected unrelated animals and from two sire families (11 animals) the Sahiwal breed Zebu cattle were investigated. Four oligonucleotide probes — (GTG)5, (TCC)5, (GT)8 (GT)12 used on genomic digested with restriction enzymes AluI, HinfI, MboI, EcoRI HaeIII in different combinations. All four produced multiloci fingerprints differing levels polymorphisms. Total bands shared each individual range 2.5 to KB. Band number ranged 9 17, 0.48 average band sharing. Probes (GT)8, (TCC)5 fingerprinting patterns medium low polymorphism, whereas probe (GTG)5 highly polymorphic patterns. Probe combination enzyme was a heterozygosity level 0.85, followed by 0.70, 0.65 0.30, respectively. GTG5 or its complementary sequence CAC5 fingerprints, indicating that can be for analysing population structure, parentage verification identifying loci controlling quantitative traits fertility status.
