摘要: Lithium and valproate (VPA) have been the most widely prescribed mood stabilizers for therapy of bipolar disorders (BD) more than 50 years. However, precise molecular mechanism their pharmacological activity is not fully known. Recent studies suggested that both drugs exert antiapoptotic effects. This review focuses on influence lithium VPA intracellular apoptotic signaling pathways. The active sites, which are implicated in mediating action, described. It has found block key proapoptotic molecules (GSK-3beta, caspase cascades) enhance survival pathways (ERK1/2 Bax proteins). potential significance reported effects discussed.
