摘要: Publisher Summary This chapter presents an analytical characterization of nucleosides in nucleic acids. The chromatography and modification is addressed from the perspective needed chromatographic methodologies for isolation acids, quantitative enzymatic hydrolysis, high resolution preparative high-performance liquid (HPLC), affinity to obtain pure single species UV absorption spectroscopy interfaced mass spectrometry investigations. experiments on determination structure-spectrum relationships, composition, conformation using array advanced techniques HPLC-UV, Fourier transform–infrared (FT–IR), nuclear magnetic resonance (NMR), spectra (MS), as well structure-RPLC retention relationships. A comprehensive discussion analytical-structural tRNAs, oligonucleotides, RNA DNA presented. Two modified nucleosides, A* G* yeast methionine initiator tRNA (initiator met ) at positions 64 65 T-Ψ stem are identified unmodified guanosine position O-β-D-ribofuranosyl-(1"→ 2')-adenosine 64.
