摘要: The present study aimed to assess the perturbation in circular RNA (circRNA)/mRNA expression profiles and a circRNA-miRNA-mRNA coexpression network involved potential protective effect of diosgenin (DIO) on alveolar bone loss rats subjected ovariectomy (OVX). Wistar (female) manipulated with sham operation were classified as SHAM group grouping OVX administered DIO, estradiol valerate or vehicle for 12 weeks was DIO group, EV respectively. Following treatments, plasmatic levels osteocalcin tumor necrosis factor-alpha microstructure assayed. Based microarray analyses, we identified differentially expressed (DE) circRNAs mRNAs both group. DE metabolism pathway validated by RT-qPCR considered key circRNAs/mRNAs. On basis these circRNAs/mRNAs, predicted overlapping relative miRNAs built. showed an anti-osteopenic rat induced OVX. In total, found 10 (6 downregulated 4 upregulated) 614 (314 300 samples compared those However, only one circRNA (rno_circRNA_016717) seven (Sfrp1, Csf1, Il1rl1, Nfatc4, Tnfrsf1a, Pik3c2g, Wnt9b) qRT-PCR therefore circRNA/mRNAs. According circRNA/mRNAs miRNAs, constructed. After analysis, axis (circRNA_016717/miR-501-5p/Sfrp1) identified. mechanism inhibiting after is possibly relevant simultaneous inhibition osteogenesis osteoclastogenesis mediating important molecules Wnt, PI3K, RANK/RANKL osteoclastogenic cytokine pathways. circRNA_016717/miR-501-5p/Sfrp1 may play roles processes.
