An Integrative View on Intra- and Inter-Cellular Cooperation Mechanisms in Alzheimer's Disease

摘要: Alzheimer’s disease (AD) has been identified as central nervous system pathology more than 100 years ago and for a long time the diagnostic criteria remained same, based on anatomopathological findings. Failure of clinical trials in almost every field AD therapy forced widening perspective molecular pathologies, to point which, now, is considered multifactorial disease. Recent advancement cell biology uncovered new data regarding amyloid precursor protein its enzymatic cleavage products, beta peptides, such oligomerization membrane pore formation. More intracellular deregulated events have highlighted, e.g. endoplasmic reticulum stress mitochondrial dysfunction, possibly related through newly discovered Mitochondria-Associated Endoplasmic Reticulum Membrane (MAM). During last few years, non-neuronal populations came into focus research, glial cells, endothelial cells forming blood-brain barrier or brain stem mesenchymal nature. All these cellular players interact react what was dogma – cascade which this about be overthrown.
