Nemo like kinase negatively regulates NF-κB activation and coelomocytes apoptosis in Apostichopus japonicus

摘要: Nuclear factor kappa B (NF-κB) transcription factors are related to several physiological processes, including innate and acquired immunity. In this study, a novel negative regulator of the Nemo-like kinase (NLK) gene was identified from Apostichopus japonicus through PCR (denoted as AjNLK). The complete AjNLK cDNA 2335 bp, with 5'-UTR 315 3'-UTR 718 putative ORF 1302 encoded polypeptide 433 amino acid residues typical serine/threonine protein domain. Blast analysis revealed that shared high degree structural conservation its counterparts other invertebrates vertebrates. Spatial expression indicated mRNA transcripts higher in tentacles than coelomocytes. coelomocytes suppressed after Vibrio splendidus challenge by 0.51-fold 0.41-fold at 72 96 h, respectively, compared control group. Similarly, down-regulated primary exposed 1 μg mL(-1) lipopolysaccharide (LPS). Functional investigation further NF-κB p105 induced both levels silencing vitro. Meanwhile, apoptosis LPS-induced significantly inhibited siRNA-transfected These results supported negatively regulated activation cell sea cucumber.
