摘要: Lung metastasis has a great influence on the prognosis of patients with osteosarcoma. We previously established two high-metastatic sublines, M112 and M132, from HuO9 human osteosarcoma cell line by in vivo selection. In this study, we newly isolated subline, H3, three low-metastatic L6, L12 L13, dilution plating method. Three sublines produced more than 200 metastatic nodules lung, while no or few after injection 2 × 106 cells into tail vein nude mice. There were significant differences motility invasiveness between high- whereas growth rates vitro tumorigenicity showed correlation their abilities. Early adherence to culture plates was significantly lower which occupied smaller surface areas other did. Comparison expression 637 cancer-related genes cDNA microarray revealed that seven differentially expressed sublines. Among them, five (AXL, TGFA, COLL7A1, WNT5A, MKK6) associated adherence, motility, and/or invasiveness. These results suggest motility/invasiveness adhesive abilities are key determinants lung
