摘要: Balanced local pH serves a vital role in extracellular microenvironment homeostasis. However, the impact of disturbance on proliferation, differentiation, and bone formation potential human adipose-derived stem cells (ASCs) remain to be fully addressed. In present study, we evaluated proliferation multilineage differentiation ability vitro by culturing ASCs mediums at 6.8 (acidic medium) 7.4 (normal medium). The results indicated that was markedly inhibited after 120 hours culture an acidic medium (p < 0.05). Additionally, osteogenic also hampered condition. Notably, adipogenic capacity unaffected. were then seeded Ultrafoam constructs induced into chondrocytes hypertrophic verify chondrogenic normal medium. condition group presented glycosaminoglycan (GAG) accumulation large chondrocyte-shaped found lacunae. contrast, showed no typical or GAG. When implanted subcutaneously mice, there de novo tissue cultured when compared blood vessel invasion group, which consistent with study. conclusion, even minor changes could significantly affect ASCs. Thus, control is crucially important successful priming, engineering, study chondrocyte physiology functions. (Am J Transl Med 2020. 5(1):25-36).
