Gender-dependent and genotype-sensitive monoaminergic changes induced by polychlorinated biphenyl 153 in the rat brain.

摘要: Polychlorinated biphenyls (PCBs) are present as ortho- and non-ortho-substituted PCBs, with most of the ortho-substituted congeners being neurotoxic. The study examined effects PCB 153 on dopamine, serotonin amino acid neurotransmitters in neostriatum both male female Wistar Kyoto (WKY) spontaneously hypertensive rat (SHR) genotypes. exposure at p8, p14 p20 had no levels these transmitters when p55, but led to increased homovanillic 5-hydroxyindoleacetic acid, degradation products dopamine serotonin, respectively, all groups except SHR. Immunoblotting showed that induced gender-specific decreases dopaminergic synaptic proteins. These included a novel finding decreased D5 receptor genders genotypes, whereas male-specific changes postsynaptic density (PSD)-95 protein WKY SHRs decrease presynaptic transporter and, less clearly A female-specific tendency vesicular monoamine transporter-2 was observed after exposure. No were seen tyrosine hydroxylase, cytoskeletal neurotubulin or plasma membrane marker Na(+)/K(+)-ATPase any strain. Hence, PCB-exposure increases transmitter turnover animals, pre- proteins predominantly animals. may therefore induce neostriatal toxicity through mechanisms genotypes genders, including aspiny interneurons, which employ mediate interneurons basal ganglia.
