Assembly of recombinant TFIID reveals differential coactivator requirements for distinct transcriptional activators.

摘要: Abstract We previously reported that transcriptional regulators can bind selected TAF subunits of the TFIID complex. However, specificity and function individual TAFs in mediating activation remained unknown. Here we report vitro assembly properties TBF-TAF complexes reconstituted from nine recombinant Drosophila TFIID. A minimal complex containing TBP II 250 directs basal but not activator-responsive transcription. By contrast, holo-TFIID supports by an assortment activators. The activator NTF-1, which binds 150, stimulates transcription with a only TBP, 250, whereas Sp1 additionally requires 110 for activation. Interestingly, 150 enhances even though this subunit does directly to Sp1.
