Complexes of tetraspanins with integrins: more than meets the eye

摘要: The transmembrane proteins of the tetraspanin superfamily are implicated in a diverse range biological phenomena, including cell motility, metastasis, proliferation and differentiation. tetraspanins associated with adhesion receptors integrin family regulate integrin-dependent migration. In cells attached to extracellular matrix, integrin-tetraspanin complexes clustered into distinct type structure at periphery. Various phosphatidylinositol 4-kinase protein kinase C isoforms, they may facilitate assembly signalling by tethering these enzymes heterodimers. At plasma membrane, partitioned specific microdomains proximal cholesterol-rich lipid rafts. A substantial fraction colocalise integrins various intracellular vesicular compartments. It is proposed that can influence migration one following mechanisms: (1) modulation signalling; (2) compartmentalisation on surface; or (3) direction trafficking recycling integrins.
