摘要: Publisher Summary A principal virtue of polyacrylamide supports in affinity chromatography is low nonspecific adsorption proteins. In cases where efficient, specific elution not feasible, this fact becomes prime importance for achieving a high separation factor. order to maintain favorable property, one should avoid loading the support unnecessarily with hydrophobic structures or charged groups. The most likely source latter would be carboxylate groups resulting from hydrolysis itself. derivatization procedures have been designed keep at level. Carboxylate can decreased by treating gel, such as (X) and presumably other amino forms, watersoluble carbodiimide (XI) presence 1 M NH 4 C1 pH 5.4. similar procedure has used purpose case Sephadex beads cellulose membranes.
