摘要: Forces have a profound role across all living systems, and many essential biological processes ranging from development migration to mitosis meiosis cannot proceed without precisely tuned mechanical signals. Despite the importance of mechanotransduction, molecular details relating magnitude, timing, location forces specific biochemical pathways remain poorly understood. The challenge pertains developing probes that allow one simultaneously measure activation tension in systems. Herein, I will describe synthesis characterization fluorescence-based turn-on fluorescence microscopy (MTFM) for imaging at lipid membrane cells. MTFM take advantage quenching as “ruler”, an extendable linker reversible “spring” with known constant. Using this technique, standard microscope can be used quantify cell In talk, second third generation high spatial temporal resolution wide range receptors types. application these sensors image associated mechano-regulatory occur cell, such endocytosis, Notch receptor activation, integrin adhesion activation.
