Brain-derived neurotrophic factor is a survival factor for cultured rat cerebellar granule neurons and protects them against glutamate-induced neurotoxicity.

摘要: We have studied the effects of different neurotrophins on survival and proliferation rat cerebellar granule cells in culture. These neurons express trkB trkC, putative neuronal receptors for brain-derived neurotrophic factor (BDNF) neurotrophin-3 (NT-3) respectively. Binding studies using iodinated BDNF NT-3 demonstrated that both bind to with a similar affinity approximately 2 x 10(-9) M. The number per cell was surprisingly high, 30 10(-4) 10(5) NT-3, Both elevated c-fos mRNA neurons, but only up-regulated encoding low-affinity neurotrophin receptor (p75). In contrast acted as neurons. also induced sprouting significantly protected them against neurotoxicity by high (1 mM) glutamate concentrations. Cultured expressed low levels which were increased kainic acid, agonist. Thus, BDNF, not is cultured activation receptor(s) up-regulates expression these cells.
