摘要: Chemical-genetic interactions (CGIs) describe a phenomenon where the effects of chemical compound (i.e., small molecule) on cell growth are dependent particular gene. CGIs can reveal important functional information about genes and also be powerful indicators compound's mechanism action. Mapping lead to discovery new probes, which, in contrast genetic perturbations, operate at level gene product (or pathway) fast-acting, tunable, reversible. The simple culture conditions required for yeast its rapid growth, as well availability complete set barcoded deletion strains, facilitate systematic mapping this organism. This process involves two basic steps: first, screening libraries identify bioactive compounds affecting and, second, measuring these genome-wide collections mutant strains. Here, we introduce protocols both steps that have great potential development small-molecule tools medicines.
