Rectal methadone in cancer patients with pain. A preliminary clinical and pharmacokinetic study.

摘要: Summary Background Cancer pain can be treated in most cases with oral analgesics. However, during their clinical history, 53% to 70% of patients will need alternative routes opioid administration. The rectal administration opioids is a simple route for many patients. There are no data the literature regarding pharmacodynamics and pharmaco-kinetics methadone Patients methods We evaluated analgesia, toler-ability absorption profile hydrochloride six opioid-naive cancer pain. A blood sample was collected before single dose drug (10 mg) then again after fixed times. At these times were asked about pain, nausea drowsiness by means visual analogue scale 0–100 mm (VAS) Results Pain relief statistically significant as early 30 minutes up eight hours None reported side effects. parmacokinetics showed rapid extensive distribution phases followed slow elimination phase Conclusions Rectal considered an effective analgesic therapy whom and/or parenteral not indicated or available
