摘要: Abstract TRPA1 receptor is activated by endogenous inflammatory mediators and exogenous pollutant molecules relevant to respiratory diseases. Previous studies have implicated as a drug target for antitussive therapy. Here we evaluated the relative efficacy of activation evoke cough. In conscious guinea pigs agonist allyl-isothiocyanate (AITC) evoked cough with maximally effective concentration 10 mM that was abolished selective antagonist AP-18. AITC (10 mM) approximately 3-times less in inducing than capsaicin (50 μM). Ex vivo single fiber extracellular recordings revealed that, similarly capsaicin, airway jugular C-fibers, but not nodose Aδ-fibers. Consistent studies, evoking sustained C-fibers. Another agonist, cinnamaldehyde, twofold more However, cinnamaldehyde (10 mM)-induced only partially inhibited AP-18, combination AP-18 TRPV1 I-RTX. We conclude naive pigs, initiates relatively modest compared initiated TRPV1, likely due lower stimulation induce
