Molecular characterization, gene expression and transcriptional regulation of thyroid hormone receptors in Senegalese sole.

摘要: Abstract Thyroid hormones (THs) play a key role in larval development, growth and metamorphosis flatfish. Their genomic effects are mediated by thyroid hormone receptors (TRs). In this study, cDNAs encoding for TRαA, TRαB, TRβ have been sequenced Senegalese sole (Soleasenegalensis). Main domains conserved motifs were identified. Also, truncated TRαB isoform (referred to as TRαBtr) spliced variant TRβv) detected. A phylogenetic analysis grouped both TRα genes into two separate clusters with their fish mammalian counterparts. Expression profiles during development juvenile tissues analyzed using real-time PCR approach. fish, TRβv, showed distinct transcript levels tissues. During metamorphosis, only TRβv modified mRNA similar way the T4 contents. To evaluate possible regulation of TRs cognate ligand larvae exposed goitrogen thiourea (TU). transcripts decreased significantly at 11 15 days after treatment. Moreover, adding exogenous TU-treated restored steady-state or even increased mRNAs respect untreated control. Overall, these results demonstrate that transcription is up-regulated THs playing major sole.
