Early midzonal oxidative stress preceding cell death in hypoperfused rat liver

摘要: Intralobular heterogeneity of oxidative stress and its topographic relationship with cell death during low-flow hypoxia were shown in perfused rat liver by digital microfluorography using dichlorofluorescin diacetate, a fluorochrome sensitive to intracellular hydroperoxide formation, propidium iodide, which labels the nuclei nonviable cells. The surface loaded two precursors was microscopically visualized, fluorescence dichlorofluorescein, highly fluorescent molecule generated hydroperoxide-mediated oxidation, digitally processed. Dichlorofluorescein significantly increased midzonal regions as early 20 minutes after starting 25% hypoxia. At 40 fluorograph showed multiple dotted patterns, intensity peaked at 60 minutes. onset studied iodide observed minutes, distribution corresponded dichlorofluorescein-enhanced regions. Allopurinol diminished retarded death. current findings show that xanthine oxidase-dependent resultant are spatially restricted intermediate zone between periportal pericentral
