Constitutive and tissue-specific differential expression of the cryIA(b) gene in transgenic rice plants conferring resistance to rice insect pest

摘要: The truncated chimeric Bt gene, cryIA(b) of Bacillus thuringiensis, driven by two constitutive promoters, 35S from CaMV and Actin-1 rice, tissue-specific pith tissue pepcarboxylase (PEPC) for green maize, was introduced into several varieties rice (indica japonica) microprojectile bombardment protoplast systems. A total 1800 putative transgenic plants could be produced. Southern analysis revealed that more than 100 independently transformed confirmed integration the gene. High levels CryIA(b) proteins were obtained in (leaves stem) many using PEPC promoter. There little difference protein level leaves stems with 35 S or Out 800 Southern-positive bioassayed, 81 showed 100% mortality insect larvae yellow stem borer (Scirpophaga incertulas). transgene, cryIA(b), different promoters a wide range expression (low to high) stably inherited number enhanced resistance. This first report indica promoter either alone combination should provide better strategy providing protection against pest resistance, minimizing seeds other tissues.
