摘要: The main limitations of microplate-based enzyme immunoassays are the prolonged incubations necessary to facilitate heterogeneous interactions, complex matrix and poorly soluble antigens, significant sample dilutions often required because presence organic extractants. This study presents use antibody immobilization on surface magnetic particles overcome these in detection mycotoxin, aflatoxin B1. Features proposed system a high degree nanoparticle dispersion methodologically simple antibodies by adsorption. Reactions between immobilized with native labeled antigens conducted solution, thereby reducing interaction period 5 min without impairing analytical outcome. Adsorption immunoglobulins nanoparticles increases their stability aqueous-organic media, thus minimizing dilution required. Testing barley maize extracts demonstrated limit B1 equal 20 pg/mL total assay duration min. Using this method, only 3-fold initial methanol/water (60/40) extraction mixture microplate wells is necessary. pseudo-homogeneous approach could be applied toward immunodetection wide range compounds.
