In Vitro Inhibition of Leishmania Attachment to Sandfly Midguts and LL-5 Cells by Divalent Metal Chelators, Anti-gp63 and Phosphoglycans

摘要: Leishmania braziliensis and infantum are the causative agents of cutaneous visceral leishmaniasis, respectively. Several aspects vector-parasite interaction involving gp63 phosphoglycans have been individually assayed in different studies. However, their role under same experimental conditions was not studied yet. Here, roles divalent metal chelators, anti-gp63 antibodies purified type I (PGs) were evaluated during vitro parasite attachment to midgut vector. Parasites treated with chelators or prior Lutzomyia longipalpis/Lutzomyia intermedia midguts sand fly LL-5 cells. In binding system used examine PG parallel. Treatment reduced Le. adhesion Lu. longipalpis midguts. The most effective compound (Phen) inhibited both vectors. Similar results observed between species cell line LL-5. Finally, parallel experiments using anti-gp63-treated parasites PG-incubated demonstrated that approaches substantially natural parasite-vector pairs infantum/Lu. braziliensis/Lu. intermedia.
