摘要: The availability of nucleotide sugars is considered as bottleneck for Leloir-glycosyltransferases mediated glycan synthesis. A breakthrough the synthesis development salvage pathway like enzyme cascades with high product yields from affordable monosaccharide substrates. In this regard, authors aim at productivities these by a repetitive batch approach. report here first time that exceptional cascade stability facilitates Uridine-5'-diphospho-α-d-galactose (UDP-Gal), Uridine-5'-diphospho-N-acetylglucosamine (UDP-GlcNAc), and Uridine-5'-diphospho-N-acetylgalactosamine (UDP-GalNAc) in multi-gram scale mode. obtained 12.8 g UDP-Gal through mass based total turnover number (TTNmass ) 494 [gproduct /genzyme ] space-time-yield (STY) 10.7 [g/L*h]. Synthesis UDP-GlcNAc mode gave 11.9 g TTNmass 522 STY 9.9 Furthermore, scale-up to 200 mL using pressure operated concentrator was demonstrated UDP-GalNAc producing resulting an 19.4 [g/L*h] 23.3 g product. conclusion, demonstrate versatile strategy stable cascades.
